House dust mite (HDM)-induced eosinophil (EOS) response is suppressed by aerosolized nicotine (aeroNic) exposure and α7 activation by highly specific α7-positive allosteric modulators (PAMs) is sufficient to inhibit the HDM response. Mice were sensitized and challenged with HDM. Bronchial alveolar lavage fluid (BALF) was collected 96 h after the last challenge with HDM. For PAM treatment, type-1 or type-2 PAMs were injected intraperitoneally just before intranasal challenges with HDM. A and B: type-2 PNU-120596 (PNU). C and D: type-1 PAM NS-1738. E and F: type-1 PAM <t>AVL-3288.</t> The results for treatment of mice with type-1 PAM NS-1738 or AVL-3288 include groups that received drug doses either 15 min or 24 h after the last challenge with HDM. In the upper panels the flow cytometry scatter plots with the accompanying marker analysis defining alveolar macrophages (AM) (CD11b+/SiglecF+) or EOS (CD11b-/SiglecF+) are shown and in the lower panel the quantitation of total BALF from up to 18 mice in each group collected from 4 independent experiments (individual mouse response is an open circle). Error bars = ± SE. AVL-3288 significantly inhibited the induction of EOS when given 15 min after final challenge with HDM.

Article Snippet: The type-1 α7 PAMs NS-1738 (Tocris, Minneapolis, MN) and AVL-3288 (MedKoo Biosciences, Morrisville, NC) were also dissolved in DMSO (10 mg/mL stock) and diluted in PBS 1:10 to deliver the same concentration (8 m/kg) used for PNU-120596.

Treatment of alveolar macrophages (AMs) in vitro with IL-4 and IL-10 activates Ccl24 by a pathway sensitive to modulation by highly specific α7-positive allosteric modulators (PAMs). A: Ccl24 transcription response of AM freshly placed in culture from bronchial alveolar lavage fluid (BALF) and treated with the cytokine(s) indicated. B: all cytokines were added at 5 ng/mL for the BALF results shown, and this value was established using a dose response as shown for IL-4/10. As shown, the minimal saturating dose of 5 ng/mL for each cytokine, respectively, was defined, and the Ki for each agent was defined. C: AM gene expression for Ccl24, IL-1β, and Ccl17, respectively, 6 h before or after the addition of IL-4/10. Measurements were made in the presence or absence of different mixtures, including nicotine and different α7 PAMs as indicated. The dashed lines reflect either the relative transcript level normalized to either the baseline IL-4/10 response (boxed treatment; upper line) or baseline expression without any treatments (lower line). Lighter gray bars identify the transcript response when IL-4/10 was coadministered with the indicated PAM without nicotine. The error bars = ± SE, and P values reflect Student’s t test outcome when paired to the control (boxed) treatment value. D: dose response for α7 PAM NS-1738 or AVL-3288 and inhibition of Ccl24 transcription induced by IL-4/10.
